Abstract
Objective: Vitamin D3 is effective in treating PCOS, however, because of its poor solvability and bioavailability, effective time is slowed and dosage requirements rise. Our previous study showed that Phytosolve containing VD3 is more effective than vitamin D3 in PCOS treatment. In the continuation, the effect of this formulation of vitamin D3 on the gene expression involved in implantation in PCOS patients was also investigated.
Material and Method: Lipid S75, glycerol, and MCT oil were combined to create the Phytosolve using a sonicator probe. Six sets of 36 female NMRI mice were created: control, sham, PCOS, Phytosolve, Phytosolve containing VD3, and vitamin D3. PCOS was induced with DHEA injections. After receiving Phytosolve containing VD3 and vitamin D3 by gavage for one week from the 13th day of model creation, female mice were mated and endometrial tissue was taken for analysis of LIF, β-integrin, and HOXA10 proteins and genes.
Results: Compared to the vitamin D3 group, the LIF, β-integrin, and HOXA10 gene’s expression in the Phytosolve-containing vitamin D3 group increased significantly (P < 0.05). The expression of proteins β-integrin and HOXA10 increased in the Phytosolve containing vitamin D3 group against the vitamin D3 group, but this increase was not significant. The increase of LIF protein in the Phytosolve containing-vitamin D3 group was significant against the vitamin D3 group (P < 0.05).
Conclusion: Vitamin D3 with Phytosolve was more successful than vitamin D3. The Phytosolve formulation can be utilized for medications with limited solubility and bioavailability.
