Pre-implantation effects of progesterone administration on ovarian angiogenesis after ovarian stimulation: a histological, hormonal, and molecular analysis

Abstract

Ovarian tissue is known as a high vascular tissue that progesterone (P4) has a direct effect on its angiogenesis. The present study was designed to show how P4 affects the ovarian angiogenesis at hormonal, histological and molecular levels. A total of 15 adult female NMRI mice were used into three groups: (control group, test group І: ovarian stimulated group, and test group П: P4 treated group after ovarian stimulation). Blood sample and ovarian tissues were used to assess hormonal, histological and molecular alterations. Gene expression was investigated for ovarian vascular endothelial growth factor (VEGF) and hypoxia inducible factor-1alpha (HIF-1ɑ) using the real time PCR. Ovarian hormones level implied an increase in the test groups compared with the control group (P<0.05). Quantitative corpus luteum parameters described an increase in the test groups compared with the control group (P<0.05). Data analysis on the quantitative ovarian vessel parameters showed a significant difference in the test groups compared to the control group. Ovarian VEGF gene expression analysis represented an increased expression in group І compared with the control group (P<0.05). No significant result was observed for HIF-1ɑ gene. P4 treatment after ovarian stimulation caused enhancement in ovarian angiogenesis via increasing hormonal levels with significant structural changes.
