P-145. In vitro sperm survival over the days following sperm capacitation

Objective: To evaluate and compare the in vitro survival of spermatozoa over three days following semen analysis with sperm capacitation.
Methods: Semen samples undergoing semen analysis with sperm capacitation were included in the study. After subjecting the samples to the discontinuous density gradient technique, the recovered spermatozoa were resuspended in 1 mL of culture medium (FertiCultTM Flushing medium, FertiPro) and incubated at room temperature (21–24°C). After the incubation periods of 24, 48 and 72 hours, samples were analyzed for sperm total motility and the % of decline in sperm survival was calculated. In order to compare sperm survival between the groups statistical analysis were performed by repeated measures ANOVA test using GraphPad Prism (version 8.0 for Windows, GraphPad Software, CA, USA) and statistical difference was considered when p value was less or equal to 0.05.
Results: Sperm survival differed statistically between the groups. Sperm survival decreased 37.17%, 64.26%, and 79.43% at 24, 48, and 72 hours respectively, compared to the immediate post-capacitation measurement.
Conclusion: The findings suggest that in vitro sperm survival after capacitation techniques declines over time. Motile sperm gradually become immotile, indicating a time-dependent reduction in motility.
