Objective: To establish the association between age and the increase of aneuploidies and DNA fragmentation rate in human sperm. To determine the age range in which there is a significant increase of aneuploidies and DNA fragmentation.To determine whether confounding variables affect the association between chronological age and the occurrence of aneuploidies and DNA fragmentation in human sperm.
Methods: This study used an observational, analytical, cross-sectional, retrospective, correlational, and comparative design. A total population of 2,930 (2017-2024) men was included, 2,680 (23-67 years old) referred infertility and 250 fertile men (21-52 years old) were recruited. Age, DNA fragmentation rate (TUNEL), aneuploidies (FISH-5 probes), seminal parameters, and smoking, fever, chronic diseases, occupation were assessed.
Results: The correlation analysis of age and sperm DNA fragmentation was R: 0.4 (p<0.05). Likewise, when the analysis was performed by grouped age, the increase of DNA fragmentation was evidenced from 45 years old (p<0.05). In men with ≥ 45 y/o lower seminal volume, progressive motility and vitality was observed (p<0.05). Contrary, aneuploidies were not significantly higher in men with more age. High level of DNA fragmentation was significantly related to low sperm count, low progressive motility, teratozoospermia, low vitality, primary infertility, recurrent miscarriage, and high risk of aneuploidies (p<0.05). Men with diabetes and commercial employee were the most related to high DNA fragmentation (54% and 33%, respectively).
Conclusion: Increase of male age linear related to sperm DNA fragmentation increase, but not the aneuploidies. Sperm DNA fragmentation increased in significantly high in men older than 45 years old. Diabetes disease and sedentary work depicted a major association with high levels of DNA fragmentation.
