Objective: To compare clinical pregnancy rates, biochemical pregnancy rates, and miscarriage rates between two groups of patients undergoing frozen single euploid embryo transfer using either Global®Total culture medium (LifeGlobal®) or EmbryoGlue® (Vitrolife). The success of in vitro fertilization (IVF) depends on multiple factors, including embryo quality, culture conditions, and the composition of the embryo transfer medium. EmbryoGlue® (Vitrolife), enriched with hyaluronan, has been suggested to enhance implantation by promoting embryo-endometrial interaction. However, evidence comparing its effectiveness with other standard culture media is still inconclusive (Bontekoe et al., 2014). This study investigates whether the type of transfer medium impacts pregnancy outcomes in frozen single euploid embryo transfers following PGT-A.
Methods: A retrospective study was conducted involving two groups of patients (n=47 each) who underwent frozen single embryo transfer with euploid embryos identified via preimplantation genetic testing for aneuploidy (PGT-A) (Igenomix®). Baseline characteristics, including recipient age, oocyte age, embryo quality and endometrial thickness were similar across both groups of study. One group received the embryo transfer using Global®Total culture medium (LifeGlobal®), while the other used EmbryoGlue® (Vitrolife). Statistical analyses were conducted using R (v4.4.1) in RStudio (v2024.12.1). The Mann-Whitney U test was applied for quantitative variables, and the chi-square test for qualitative variables when comparing the Global total and Embryoglue groups. Quantitative results are presented as mean±standard deviation, and qualitative variables as counts and percentages of patients who received single embryo transfer.
Results: There were no statistically significant differences between the two groups in terms of clinical pregnancy rate, biochemical pregnancy rate, or miscarriage rate. A non-significant trend toward higher clinical pregnancy rates was observed in the group using Global®Total culture medium (LifeGlobal®). However, this trend did not reach statistical significance, likely due to the limited sample size.
Conclusion: The use of either Global®Total culture medium (LifeGlobal®) or EmbryoGlue® (Vitrolife) in frozen single euploid embryo transfers results in comparable outcomes regarding pregnancy and miscarriage rates. While a slight trend favoring the global medium was observed, the differences were not statistically significant. Larger prospective studies are needed to determine whether these trends hold true in larger populationss.
