Folliculogenesis in transplanted rat ovaries after freezing/thawing

Abstract

Objective: Evaluate the capacity of transplanted rat ovaries to respond to ovulation induction with follicular stimulation hormone, after freezing/thawing. Methods: A total of 30 Holtzman rats at the same age, were included in the study and split in 3 groups. In group I ovulation induction with FSH was performed after Freezing/Thawing and transplantation of the ovary to the peritoneum; in group II ovulation induction with FSH was performed after transplantation of the ovary to the peritoneum; in group III, ovulation induction with FSH was performed after laparotomy was performed (shame operation). Slow freezing was performed in all groups using DMSO and thawing was performed at room temperature at the day transplantation was performed. Ovulation induction was performed on day 15 after surgery. Ovaries were removed on day 4 of the oestral cycle for histological analyses. Results: Ovaries of group I had four primary follicles, eight pre-antral follicles and 13 antral follicles; group II, seven primary follicles, 11 pre-antral follicles and 10 antral follicles; and group III one primary follicle, 14 preantral follicles and 16 antral follicles. Frozen/thawed and transplanted ovaries did respond to ovulation induction with FSH similar to normal and non-frozen ovaries. Conclusion: Cryopreservation, thawing and transplantation of rat ovarian tissue, with no vascular anastomosis, to the peritoneum, did not jeopardize follicular development after ovulation induction with follicular stimulation hormone.
