Mus domesticus domesticus in vitro embryo culture from one cell to ecloded blastocist in KSOM and HTF medium modified

Abstract

The experiments were design to determine the viability of a mice embryo in vitro culture system, using RTF and KSOM media, as a quality control of an human in vitro fertilization programo Experiment 1 : embryos 1- cell (CF1xSWISS) were cultivated in KSOM and RTF supplemented with 20% of bovine fetal serum. Experiment 2: observation of the cleavage after addition of bovine serum albumin to the culture medium. Experiment 3: determination of the cleavage and hatched blastocyst rate in the presence of the bovine serum albumin and REPES in the culture media. The percentage of development to 2-cells embryo was 73.6% with KSOM and 58.5% with RTF in the first experiment. The KSOM supplemented with bovine serum albumin result in similar percentage of cleavage compare with the first experiment. ln the third experiment the percentage of development to 2 cells embryo was 84,9% with KSOM and 84.2% with RTF. The percentage of development to the hatched blastocyst stage was 58,5% with KSOM and 56.2% with RTF. The RTF and KSOM media supplemented with BSA and REPES are able to support Mus domesticus domesticus embryo development to the hatched blastocyst stage. This system may be utilized as a quality control of RTF medium utilized in human in vitro fertilization procedures
