Embryo development after ICSI into zona-free oocyte

Abstract

Introduction: To show that zona-free oocytes can be normally fertilized with the use of intracytoplasmatic sperm injection (ICSI). Material and Methods: Patient l.C.N.R.A., 27 years old, submitted to the first assisted fertilization cycIe due to the presence of severe male factor and polycystic ovary syndrome (PCOS) . A total of 20 oocytes were retrieved, of which 8 MII were subjected to removal of cumulus cells with hyaluronidase for the ICSI realization. One of these oocytes escaped from the zona pellucida and was also subjected to ICSI. Results: Ali the 8 oocytes which were subjected to ICSI were fertilized. The oocyte without zona pellucida cleaved on day 2 (2cells), presenting normal morphology (grade 1) . On day 3 the pre-embryo presented 4 cells (grade 1), on day 4 it presented 4 cells (grade 1), on day 5 it presented 8 cells (grade 1) and on day 6 it presented degeneration, without reaching the blastocyst stage. Among the other 7 embryos, 4 of them reached the blatocyst stage and were transferred, resulting in pregnancy. Conclusion: This case demonstrates that zona-free oocytes can undergo normal cleavage until the 8 cell stage.
