Abstract
Introduction:To compare assisted reproductive techiques(ART) results using two different routes of administration of GnRH analogues(daily vs depot).
Materiais and Methods:During the period July 1995 until May 1996 one hundred cycles were randomized in two groups at the time of admission in our ART program. All patients received GnRH analogues on day 21st of the cycle. Group I received a single intramuscular injection of 3.75 mg of Leuprolide Acetate (LA) (Lupron Depot 3.75mg) and group II received daily subcutaneous administration of 1mg of LA(Lupron 2.8 ml) until hCG administration day. Gonadotropin ovarian stimulation(hMG-FSH) began on cycle day 3.
Results:There were no significant differences in the number of ampoules of gonadotropins used in each group (33.6±9.4 vs 30.3±4.8) nor in the aspiration day (15.1±1.7 vs 14.7±1.2). E2 leveis on hCG day were 1458±1140pg/ml in group I and 2036±1247pg/ml in group II (p<0.02). There was no fertilization in one case in group I and in three cases in group II; besides in one patient in group II no spermatozoa were found in the testicular biopsy.Therefore, 39 transfers were done in group I and 33 in group II. There were no significant differences in the total(30.7 % vs 24.2%), clinical(23.1 % vs 21.2%) and in progress(20.5% vs 18.1%) pregnancy rates. The implantation rate was similar in both groups(9.4% vs 7.1% ).
Conclusion:Daily and depot administration of GNRH analogues for ovarian suppression prior to ovarian stimulation for ART showed similar results. Since depot administration is more comfortable and convenient for the patient this route seems to be preferable.
